Propriétés

  • Antalgique [2] (action sur les récepteurs muscariniques, opioïdes et dopaminergiques) [3], [4], [5], action sur les récepteurs N-Methyl-d-aspartate (NMDA) [6], action sur les récepteurs à glutamate [7]
  • Anti-inflammatoire [8], inhibe l'œdème induit par la carragénine [9]
  • Hypotenseur, bradycardisant [10]
  • Sédatif, calmant, hypnotique, anticonvulsivant
  • Le linalol est neuroprotecteur dans un modèle in vitro de stress oxydatif induit par le glutamate, potentialités dans les maladies cérébrales neurodégénératives [11]
  • Anxiolytique [12]
    • Inhibe la liaison du glutamate dans le cortex cérébral [13], [14], interfère avec la transmission glutamatergique [15], supprime la fonction des récepteurs excitateurs du glutamate, inhibition non compétitive dose-dépendante de la liaison de [(3) H] MK801 (antagoniste du NMDA) [16]
    • Affinité pour les récepteurs au GABA-A (métabolites oxygénés au carbone 8) [17], que d'autres travaux ne retrouvent pas chez le rat [18]
    • Inhibe la libération d’acétylcholine et réduit le temps d’ouverture des canaux ioniques de la jonction neuro-musculaire (blocage des canaux Na+ et/ou Ca ++)
  • L’inhalation d’odeurs de jasmin et de lavande induisent une diminution significative de la fréquence cardiaque, effet attribué au (R)-(-)-linalol [19]
  • Spasmolytique sur les muscles lisses intestinaux (surtout) [20], [21] et trachéaux [22], par un mécanisme vraisemblable de stimulation de l'enzyme adénylate cyclase, produisant une augmentation de l'AMPc, le linalol inhibe la libération d'ACh au niveau de la jonction neuromusculaire de souris [23]
  • Inhibe la libération d'ACTH, d'adrénaline et de gonadotrophines chez la rate [24]
  • Antioxydant (inhibe la peroxydation lipidique), hypothermisant
  • Anesthésiant local
  • Antibactérien [25]
  • Antifongique, actif contre des souches de Candida résistantes au fluconazole [29]
  • Antiviral, le linalol montre une forte activité contre les Adenovirus-2 (AVD-II), responsables de pharyngites, pneumonies et gastro-entérites [30]
  • Antitumoral, stimule diverses cytokines : IFN-γ, IL-13, IL-2, IL-21, IL-21R, IL-4, IL-6sR, TNF-α, induit la réponse immunitaire de type Th1 [31], inducteur d’apoptose [32]
  • Attractif pour les pollinisateurs
  • Il possède l'odeur du muguet

Effet thérapeutique

Effets indésirables

  • Potentiellement allergisant :
    • L'une des 26 substances allergènes dont la présence doit être mentionnée dès lors que leurs concentrations dépassent :
      • 0,001% pour les produits à ne pas enlever (crèmes)
      • 0,01% pour les produits à enlever par rinçage (Directive européenne, 2003)
    • Ce sont surtout les produits d’auto-oxydation du linalol qui sont allergisants [33]
  • Peu toxique :
    • DL50 par voie orale chez le rat : 2 790 mg/kg, DL50 par voie cutanée chez le lapin : 5 610 mg/kg [34]
  • Métabolisé par glucuronoconjugaison puis excrété dans les urines [35]
  • Attention à d'éventuels effets gynécomastiants eu usage prolongé, linalol et acétate de linalyle montrent une activité anti-androgénique et une faible activité œstrogénique en se liant aux récepteurs aux œstrogènes [36]
