Resvératrol = 3, 4′, 5 trihydroxystilbène

Famille moléculaire

Polyphénol de la famille des stilbènes

  • Phytoalexine = substance induite par un stress environnemental ou pathogène (par exemple une attaque fongique)
    • Sa présence dans le fruit de la vigne rouge provient d’une réaction [1] contre une infection due au champignon Botrytis cinerea
    • Il sera logiquement plus abondant dans des raisins non traités
  • L'isomère trans-resvératrol est plus abondant dans les plantes que la forme cis- (sous forme glycosylée, trans-resvératrol-3-O-glucoside). Son activité anti-oxydante serait 7 fois supérieure à celle du cis-resvératrol

Source végétale

  • Pellicule du grain de raisin de la vigne rouge et vin rouge (surtout le Pinot noir, teneur moyenne en France 5,4 ± 1,2 mg/L) [5]
  • Fèves de cacao et chocolat noir (0,4 mg/kg de resvératrol)
  • Baies rouges du genre Vaccinium : jus de canneberge (0,278 mg/kg)
  • Arachide (cacahuètes 0,03 à 0,147 mg/kg)
  • Rhubarbe, mûre, grenade, sorgho
  • Renouée du Japon Fallopia japonica (Houtt.) Ronse Decr. = Reynoutria japonica = Polygonum cuspidatum dont la racine, médicinale en Chine et au Japon, est riche en resvératrol (2,96-3,77 g/kg)
    • La plante est consommée en infusion (« thé d’itadori », traditionnellement utilisé dans le traitement des maladies cardio-vasculaires et comme anti-inflammatoire)
    • Elle devient particulièrement invasive en Europe et en Amérique, surtout sur des sols naturellement riches en composés métalliques (au Japon, c'est une espèce pionnière des pentes de volcans)
  • Le mûrier blanc Morus alba Moraceae contient l'oxyresvératrol, puissamment anti-inflammatoire et imunomodulant [2]

Propriétés

  • Protecteur d'enzymes impliqués dans l'excrétion des xénobiotiques par l'organisme
  • Antagoniste compétitif du Récepteur des Arylhydrocarbures (AhR), qui se lie aux dioxines, à la fumée de tabac, aux particules diesel, aux PCB, aux hydrocarbures, permettant une détoxication spécifique [3], [4]
  • Le resvératrol améliore l’espérance de vie par activation des sirtuines et des molécules SIRT1 [5]
  • Antioxydant, diminution de l’expression des cytokines IL-1b, IL-6, IL-8
  • Antagoniste des voies NF-kB et AP-1
  • Inhibe la prolifération des cellules musculaires lisses vasculaires [6], vasoprotecteur, atténue le stress oxydatif mitochondrial [6], [7]
  • Inhibe la carcinogenèse aux stades d'initiation, de promotion et de progression
    • C'est l'une des nombreuses substances naturelles qui sont impliquées dans la prévention de la cancérisation [8]
  • Protection vis-à-vis de la prolifération de cellules de cancer du foie [9]
  • Puissant inhibiteur de la progression des tumeurs du sein, de la peau et du colon [10], [11], de l'ovaire [12], prévention du cancer du colon [13]
  • Association intéressante avec les chimiothérapies par action sur les RAFT et les protéines Bcl2 (antiapoptotiques) qui réduisent l'efficacité des antitumoraux [14], [15], [16]
  • Prévention de la toxicité cardiaque de la doxorubicine, avec quercétine [17]
  • Certaines études sont contradictoires (cancer du sein) [18]
  • Anti-inflammatoire [19], impliqué dans la suppression de l'inflammation et la prévention des cancers par inhibition probable de l'activation de NF-kB et enréduisant l'activation de IKK. [20] [7], inhibition des cyclooxygénases (COX-1 et COX-2) => modification de la synthèse des éicosanoïdes, le resvératrol inhibe la production d'IL-1β, MMP-3 induite par le TNF-α dans les synoviocytes de la polyarthrite rhumatoïde humaine, intérêt dans la polyarthrite rhumatoïde [21]
  • Agent de chimio-prévention du cancer, propriétés anti-inflammatoires, neuroprotectrices et antivirales [22]
  • Anticancer, module l’expression de micro-ARN (séquences non codantes de l’ARN) impliqués dans la réponse inflammatoire et dans la naissance des cancers par une nouvelle voie de signalisation [23], anti-métastatique [24], inducteur d'apoptose et rôle potentiel dans la sensibilisation à la radiothérapie du mélanome [25], de la prostate [26]
  • Inhibe la prise de poids chez des primates non-humains (lémurien microcèbe) [27], [28], améliore la survie de la santé de souris au régime hypercalorique [29]

  • Le resvératrol posséderait la capacité de prolonger les années de vie en bonne santé et de limiter les maladies liées à l'âge [52], de façon différente selon les espèces [53], et dans beaucoup d’espèces, le régime alimentaire et l’apport calorique jouent un rôle prépondérant lors de l’association avec la prise de resvératrol alimentaire [54], [55]
  • Effet prébiotique favorable sur la flore intestinale
  • Le resvératrol est une molécule prometteuse dans la lutte contre les résistances bactériennes, il potentialise l'activité de la colistine contre un large panel de souches résistantes [56], quelles que soient les espèces et les mécanismes de résistance, et sans montrer intrinsèquement d’activité antimicrobienne. Il améliore l'efficacité des aminoglycosides contre Staphylococcus aureus [57] et de la polymyxine B sur Klebsiella pneumoniae et Escherichia coli [58]
  • Mais sa demi-vie d'élimination est très courte et sa biodisponibilité faible, la galénique est très importante

  • Très bonne sécurité d'emploi [63]
  • Pas de nocivité connue [64] à la dose de 150 mg/jour chez l'homme [65], jusqu'à 300 mg/kg/jour chez le rat [66], [67]
  • Interaction avec les cytochromes P450 1A2 (inhibition)
